Clearer Profiles for Confident Mixture Interpretation
The Paradyme™ 27GY STR System provides all of the materials needed for co-amplification and 8-color fluorescent detection of 27 loci, including the expanded CODIS core loci, three Y-STR loci (DYS391, DYS570 and DYS576) and Amelogenin for gender determination. The 8-dye chemistry enables shorter amplicons, with all loci below 400bp, increasing the likelihood of generating informative profiles from degraded samples.
The system incorporates Reduced-Stutter Polymerase (RSP) that limits stutter artifact formation before interpretation begins. Clearer electropherograms simplify both human review and software-driven mixture analysis:
- Determination of the Number of Contributors (NoC) is faster and more accurate.
- Minor contributor alleles in the stutter positions of a major profile become visible.
- Interpretation is more consistent from analyst to analyst.
Two internal quality indicators, QIS (small) and QIL (large), provide sample-level assessment of amplification performance, helping analysts distinguish between inhibited and degraded samples to choose the right path forward without unnecessary rework.

In a comparison study of the Paradyme™ 27GY STR System and a non-RSP 6-dye STR kit, six former DNA analysts analyzed 20 mixtures of up to five contributors. Analysts had varying levels of experience (5–20 years) and were blinded to the number of contributors. Across all analysts and samples, Paradyme™ 27GY STR System demonstrated higher interpretation accuracy, reduction in interpretation time, and higher analyst confidence.

Excellent Sensitivity for Low-Template Samples
The Paradyme™ 27GY STR System reliably produces complete STR profiles from low amounts of template DNA, calling 100% of expected alleles down to 62pg and approximately 95% of alleles at 31pg.
Seven replicate PCR amplifications of 1.0ng, 0.5ng, 0.25ng, 0.125ng, 0.0625ng, 0.03125ng and 0.015625ng purified human DNA were amplified on a ProFlex thermal cycler using the recommended cycling protocol. Amplification products were electrophoresed on a Spectrum CE System. Percent allele calls at each template amount is plotted on the Y axis.
Reduced Stutter Makes Probabilistic Genotyping Better
Probabilistic genotyping (PG) software does not remove stutter; it models expected stutter behavior at each locus and weighs whether each small peak is stutter or a true allele. Every ambiguous peak weakens the resulting statistics. Because RSP reduces stutter before interpretation, less stutter needs to be modeled probabilistically—cleaner data in, better statistics out.
PG results are also more accurate when the assumed contributor count is correct—Paradyme™ 27GY STR System demonstrated substantially more accurate NoC determination in a blind evaluation study.
Developed for Spectrum CE Systems
Promega’s 8-dye chemistry was developed for the Spectrum CE family, with a size for any laboratory throughput:
- Spectrum Compact CE System (4 capillaries, up to 32 samples)
- Spectrum CE System, 8-Capillary (>1,500 samples per week)
- Spectrum CE System, 24-Capillary (>4,000 samples per week)
Eight-dye data can be analyzed with GeneMarker® HID Software for Spectrum CE Systems and FaSTR™ DNA software (PHF Sciences).
