Quantifique a Liberação de Citocinas sem as Tediosas Etapas de Lavagem
O imunoensaio Lumit™ IFN-γ mede quantitativamente o IFN-γ liberado em amostras de cultura de células usando um protocolo simples e sem lavagem. Basta adicionar anticorpos marcados à amostra, adicionar o reagente de detecção e ler o sinal luminescente usando um luminômetro de leitura de placa padrão. Todo o protocolo é concluído em menos de 70 minutos! O ensaio pode ser usado diretamente nas células em cultura ou no meio de cultura transferido para uma placa de ensaio separada.
Como Funciona o Imunoensaio Lumit™ IFN-γ
Os anticorpos primários para IFN-γ foram selecionados por sua detecção específica e sensível e marcados com as subunidades LgBiT e SmBiT da NanoBiT® Luciferase. Na presença de IFN-γ, as subunidades são reunidas para formar uma enzima luciferase ativa. A adição de substrato otimizado gera um sinal luminescente brilhante proporcional aos níveis de IFN-γ.

Detectar IFN-γ como um Marcador de Ativação de Células T
Purified CD8+ T cells (effector cells) were combined with Raji B cells (target cells) and a serial dilution of Blincyto® (CD3 and CD19 bispecific T cell engager). The Lumit® IFN-γ (Human) Immunoassay reagents were added directly to treated cell wells to detect IFN-γ release from the T cells.

Human PBMCs were plated in a 96-well plate in 100µl and treated for 24 hours with cell stimulation cocktail (CSC), lipopolysaccharide (LPS) or R848. Four 20μl aliquots were transferred to a 384-well plate for detection of IL-2, IL-6, IFN-γ and TNF-α with the corresponding Lumit® Cytokine Immunoassay. This simple approach for profiling cytokine release can be applied to multiple cell models.
