Bio-Glo-NB™ VLP Luciferase System Assay Principle

The Bio-Glo-NB™ VLP Luciferase Assay Systems are functionally tested for performance using HiBiT-PsVLP Bioassays and intended for use with those products. When pseudotyped, HiBiT-tagged VLPs are added to target cells expressing the relevant receptors, and the presence of inhibitors or neutralizing antibodies (nAbs) prevents the entry/fusion process of the PsVLPs, resulting in HiBiT not being released intracellularly, and no luminescent signal is produced. In the absence of inhibition, the HiBiT-PsVLPs bind to the target cells and undergo membrane fusion. HiBiT is released into the target cells to bind LgBiT and generate a luminescent signal.

Assay Workflow

The Bio-Glo-NB™ VLP Luciferase Assay is a nonlytic detection system designed to measure intracellular NanoBiT® complementation and luminescence in living cells. This system is intended for use with our quantitative mechanism-of-action (MoA)-based Reporter Bioassays. The cell impermeable Bio-Glo-NB™ DrkBiT peptide is pre-incubated with cells to prohibit extracellular NanoBiT® complementation and eliminate extracellular background. The Bio-Glo-NB™ Live-Cell Reagent contains the cell permeable substrate, furimazine, and a proprietary agent for reducing autoluminescence and decreasing assay background.
